• Monotonic– e.g. For every MFI, there is only one
concentration

Assumptions of the 4PL & 5PL
• 2 assumptions about how the data are scattered around the curve…1. The scatter follows a Gaussian or “normal”
distribution
2. The standard deviation of the scatter (of the response values or MFI) is the same for all values of x (or concentrations in our case)

Weighting
• 1/Y^2 (Relative weighting) – Most commonly used
• 1/Y – Most effective when the scatter follows a Poisson distribution
• Mainly affects points on the lower portion of the curve
• Test by doing before/after on the % Recovery
