ANTI CARDIOLIPIN ANTIBODY

ACA is form of antibody directed against CARDIOLIPIN.
Cardiolipin is an important component of the inner
mitochondrial membrane, where it constitutes about 20% of
the total lipid composition.
when immune system recognize cardiolipin as a foreign
substance, ACA is produced and results in auto immune
disorder.
WHAT IS ACA?
Manual method for ACA test
The ACA test detects the autoantibodies
present in an individual’s blood serum. The common test used for detecting and
quantifying ACAs is ENZYME LINKED IMMUNOSORBANT ASSAY(ELISA).
1st step :
Sample addition – 490µl diluent + 10µl sample
Incubation – 30 minutes at room temperature.
Washing – 3 times with wash buffer either manually or automatically.
2nd step :
Addition of conjugate – 100µl conjugate
Incubation – 30 minutes at room temperature.
Washing – washed with wash buffer.
3rd step :Adding TMB solution – 100 µl substrate
Adding stop solution - Stopping the
enzymatic reaction.
MEASUREMENT
Qualitative results provide a simple positive or negative result.
In quantitative ELISA, measured at 420nm in colorimeter.
oAnti cardiolipin antibodies are found in several diseases,
including syphilis, antiphospholipid syndrome, livedoid
vasculitis, vertebrobasilar insufficiency, and Systemic
Lupus Erythematosus(SLE).
oThey are form of anti-mitochondrial antibody.
o In SLE, anti-DNA antibodies and anti-cardiolipin
antibodies may be present individually or together
